A repication-competent recombinant poliovirus which expresses an exogenous protein as a comp of recombinant polyprotein precursor that is proteolytically processed, with the result that the exogenous protein is released from the said precursor, in which the recombinant genome comprises - (A) An exogenous nucleic acid seq encoding an exoge nous protein to be expressed - (B) An exogenous seq which is a polioviral proteolytic cleavage site & which is cleaved by poliovirus protease, wherein the protease proteolytically processes a protein precursor produced by parent poliovirus modified to make the recombinant poliovirus - (C) The genome of the parent poliovirus modified to make the recombinant poliovirus Wherein (A) & (B) are inserted in (C) at a location in the genome of the parent poliovirus such that they do not disrupt a polioviral seq necessary for polioviral replication & when expressed, the exogenous protein is expressed as a comp of the polyprotein precursor produced by the recombinant poliovirus, the said precursor is proteolytically processed by the recombinant poliovirus & the exogenous protein is not included in a mature poliovirus particle produced by the replication-competent poliovirus |
