-- Protease
- Recovering isolated basal stem cells using soln
-- (I) at concn 16 - 22 mM
-- NaCl (90 - 140 mM)
-- Protease inhibitor
- Culturing said isolated basal stem cells in a medium to form
a confluent sheet of undifferentiated tissue, said medium
comprising
-- (I) at concn 14 - 22 mM
-- NaCl (100 - 120 mM)
-- Histidine (0.1 - 0.25 mM)
-- Isoleucine (0.05 - 0.5 mM)
-- Methionine (0.1 - 0.5 mM)
-- Phenylalanine (0.1 - 0.5 mM)
-- Tryptophan (0.05 - 0.5 mM)
-- Tyrosine (0.1 - 0.5 mM)
-- Insulin-like growth factor -1 (0.3 - 30 ng/ml)
- Culturing said sheet of undifferentiated tissue in a differentia
tion medium to form a sheet of undifferentiated & stratified
tissue using a medium comprising
-- (I) at concn 14 - 22 mM
-- NaCl (100 - 120 mM)
-- Ca(II) ions (0.7 - 3 mM)
-- Histidine (0.1 - 0.25 mM)
-- Isoleucine (0.05 - 0.5 mM)
-- Methionine (0.1 - 0.5 mM)
-- Phenylalanine (0.1 - 0.5 mM)
- Tryptophan (0.05 - 0.5 mM)
-- Tyrosine (0.1 - 0.5 mM)
-- Beta transforming growth factor (3 - 30 ng/ml)
